Triveni Bio, a Watertown, Massachusetts-based clinical-stage biotechnology company, has raised $65 million in Series C funding co-led by Ascenta Capital and Janus Henderson Investors. The company develops first-in-class therapeutics targeting root causes of immunological and inflammatory disorders using a genetics-informed approach. The capital will expand clinical studies for its bispecific antibody TRIV-573 and support next-stage growth.
Genetics Approach Challenges Inflammation Focus
The timing aligns with growing recognition that current therapies leave substantial unmet need in atopic dermatitis. Up to two-thirds of patients continue to struggle with uncontrolled disease despite existing treatments. Triveni targets skin barrier dysfunction through KLK5/7 inhibition rather than solely downstream inflammation pathways.
Uncontrolled Disease Persists for Most Patients
Atopic dermatitis affects millions, yet the company estimates roughly 60 percent of moderate-to-severe patients fail to achieve adequate control on approved therapies. Existing treatments primarily address Th2 inflammation while leaving barrier dysfunction and itch unresolved in many cases. This gap drives interest in upstream mechanisms validated by human genetics.
Dual KLK Inhibition Addresses Barrier Root Cause
Triveni’s lead candidate TRIV-509 is a half-life extended monoclonal antibody that dually inhibits kallikreins 5 and 7. It simultaneously targets barrier dysfunction, inflammation, and itch. The follow-on bispecific TRIV-573 adds IL-13 blockade to the KLK5/7 mechanism. Both programs are now in the clinic, with Phase 2 data for the lead asset expected by the end of 2026.

Ascenta Capital and Janus Henderson Investors co-led the round, with participation from Deep Track Capital and existing backers. Janus Henderson’s involvement as a public-market asset manager alongside specialist biotech funds suggests conviction in both near-term clinical catalysts and longer-term strategic optionality.
Immunology Market Expands on Novel Mechanisms
The global immunology market stood at $123.05 billion in 2026 and is projected to reach $228.18 billion by 2034. Within atopic dermatitis specifically, the market is expected to grow from $13.1 billion in 2025 to $22.4 billion by 2033. Capital continues flowing toward programs that move beyond established IL-4/IL-13 pathways toward genetics-validated targets with potential to improve response rates.
Triveni Bio emerged from the 2023 combination of Amagma Therapeutics and Modify Therapeutics and has now raised approximately $272 million across three rounds.
